The present invention includes compositions and methods for treating diseases or disorders associated with pathological calcification or pathological ossification. In certain embodiments, the diseases or disorders are selected from the group consisting of Generalized Arterial Calcification of Infancy (GACI), Idiopathic Infantile Arterial Calcification (IIAC), Ossification of the Posterior Longitudinal Ligament (OPLL), hypophosphatemic rickets, osteoarthritis, calcification of atherosclerotic plaques, PXE, hereditary and non-hereditary forms of osteoarthritis, ankylosing spondylitis, hardening of the arteries occurring with aging, calciphylaxis resulting from end stage renal disease and progeria.

SELF-REGULATING AAV VECTORS FOR SAFE EXPRESSION OF MECP2 IN RETT SYNDROME

In some aspects, the disclosure relates to compositions and methods of engineering a transgene. In some embodiments, the disclosure provides self-regulating recombinant nucleic acids, viral vectors and pharmaceutical compositions comprising a MeCP2 transgene. In some embodiments, compositions and methods described by the disclosure are useful for treating diseases and disorders associated with a loss of function mutation, for example Rett syndrome.

TISSUE SELECTIVE TRANSGENE EXPRESSION

Provided herein are compositions and methods for selective expression of a transgene. Compositions and methods for selective expression of a transgene comprise one or more human regulatory elements, which, when operably linked to a transgene, can facilitate selective expression of a transgene (e.g., cell-type selective expression) in a target cell as compared to at least one or more non-target cells.

EXPRESSION VECTORS AND RELATED METHODS OF DELIVERY OF Na/K ATPASE/Src RECEPTOR COMPLEX ANTAGONISTS

Expression vectors are provided that comprise a nucleic acid sequence encoding a polypeptide antagonist of a Na/K ATPase/Src receptor complex. The nucleic acid encoding the polypeptide antagonist is operatively linked to a promoter for expressing the polypeptide antagonist in a specific cell or tissue. Viral particles, target cells, and pharmaceutical compositions are also provided and include the expression vectors. Methods of treating a Src-associated disease is further provided and includes administering the expression vectors encoding the polypeptide antagonist of the Na/K ATPase/Src receptor complex to a subject in need thereof.

COMPOSITIONS AND METHODS FOR TREATING SENSORINEURAL HEARING LOSS USING OTOFERLIN DUAL VECTOR SYSTEMS

The disclosure features compositions and methods for the treatment of sensorineural hearing loss and auditory neuropathy, particularly forms of the disease that are associated with mutations in otoferlin (OTOF), by way of OTOF gene therapy. The disclosure provides a variety of compositions that include a first nucleic acid vector that contains a polynucleotide encoding an N-terminal portion of an OTOF protein and a second nucleic acid vector that contains a polynucleotide encoding a C-terminal portion of an OTOF protein. These vectors can be used to increase the expression of OTOF in a subject, such as a human subject suffering from sensorineural hearing loss.

Two or more conditional, dominant, lethal gene expression systems provide high levels of penetrance in insects. Lethality is induced at an earlier stage of development and the risk of biochemical resistance is reduced, as compared to a single insect conditional, dominant, lethal gene expression system. The invention is useful for the control of insect populations.

COMPOSITIONS AND METHODS OF DUAL POISON-ANTIDOTE MEIOTIC DRIVERS

The present disclosure provides, inter alia, meiotic drive genes derived from chromosome 3 of the fission yeasts Schizosaccharomyces kambucha and S. pombe and orthologs thereof. In certain embodiments, compositions, methods, and kits are provided for biasing organisms to express such genes and, optionally, to co-express one or more genes of interest in such organisms.
